This element is used to transmit light through a fiber optic cable from a light source (illuminator) to a microscope XY stage. Such a stage is then used for transmitted light observation. A setup consisting of a light source - fiber optic light ring - XY stage was used for example in Leica DVM systems or in motorized Märzhäuser / Prior stages used in many microscopes, for example the Leica DM6000 microscope. This is a fiber optic light ring for a stage - not for one specific microscope, because such an illuminated stage can be mounted to various microscopes.

microscope element allows expansion of a transmitted light microscope with a module for reflected light (EPI) observation:

because this is exactly a "universal" illuminator.

The set includes a reflector cube for reflected light brightfield work. This is the Leica 1 BF 561034 cube (alternative number format: Leica 561 034).

Basic technical parameters of the Leica 565015 universal EPI reflected light illuminator:
- 4 filter cubes (reflectors) can be installed, for example brightfield BF (already installed here), darkfield DF, fluorescence cubes
- 4 round filters with 32 mm diameter can be installed
- a light source must be installed
- a polarizer and analyzer can be installed
- mounting connection to microscope body: 44.5 mm dovetail
- head mounting connection: 44.5 mm dovetail

The Leica 565015 EPI illuminator is modular and can be configured according to individual requirements. The example shown and offered here is a basic version which must be completed according to the user's own needs (however the "Leica 1 BF" reflector is already installed).

NOTE - as visible in the photos and in the video, the cover flap (protective cover) which covers the rotating turret with filter cubes (reflectors) is missing.

Important for certain applications, this illuminator features passive cooling, meaning a heat sink instead of a fan. As a result, there are no vibrations that are often transferred to the workbench and then to the microscope specimen. This is particularly important when observing in-vivo specimens where the medium is a droplet of water. In such cases, vibrations generated by a fan can cause movement of objects (for example microorganisms) within the water droplet, resulting in a blurred image.

IMPORTANT: this listing includes only the light source - no fibre optic light guide or ring illuminator is included.

This illuminator provides intense green fluorescence illumination for:

Light at 530 nm is commonly used in two optical configurations.

1. As excitation light (Excitation). It excites fluorescent dyes with yellow, orange and red emission spectra:
- Cy3 / Alexa Fluor 546 / Alexa Fluor 555 - widely used markers in immunofluorescence
- TRITC / Rhodamine - classic rhodamine dyes used for antibody and cytoskeleton (actin) labelling
- dTomato / mStrawberry - red fluorescent proteins used in in vivo studies

2. As an emission detection band (Emission). In the classic blue-light excitation configuration (for example with a 488 nm laser), 530 nm corresponds to the emission maximum (green fluorescence) of:
- GFP (Green Fluorescent Protein) - a standard marker in genetic engineering and cell biology
- FITC / Alexa Fluor 488 - the most commonly used dyes for labelling cellular structures
- Fluo-4 / Calcein - indicators used to measure calcium ion concentration and cell viability

This illuminator does not use any unusual microscope mounting method - it employs a standard 48 mm dovetail mount. As a result, it may be possible to adapt it to other Leitz or Leica microscopes and even to microscopes manufactured by other companies.
